       双光子吸收/激发技术的原理:在强光激发下,分子同时吸收两个光子,从基态跃迁到两倍光子能量的激发态的过程。两个光子可以是相同波长的,也可以是不同波长的,但必须是同时吸收(两个光子到达被激发分子的时间间隔小于1飞秒)。可以这样理解,先吸收一个光子的能量跃迁到一个虚拟的中间态,然后再吸收一个光子的能量跃迁到激发态。 双光子显微镜相对于共聚焦等其他显微镜的优点: ~ 光损伤小:由于双光子显微镜使用的是可见光或近红外光作为激发光源,这一波段的光对活体细胞和组织的光损伤小,适用于长时间的研究; ~ 穿透能力强:相对于紫外光,可见光和近红外光都具有更强的穿透能力,因而受生物组织散射的影响更小,解决对生物组织中深层物质的层析成像研究问题;研究表明,共聚焦荧光成像的成像深度一般在50微米左右,而双光子显微镜的成像深度可达1000微米; ~ 高分辨率:由于双光子吸收截面很小,只有在焦平面很小的区域内可以激发出荧光,双光子吸收仅局限于焦点处的体积约为波长3次方的范围内
